45 Catálogo de Investigaciones | Pontificia Universidad Católica de Puerto Rico Identification of Cytochrome P450 3A4 and 2C9 present in Cho Cells treated with Morinda citrifolia The Morinda citrifolia, also known as Noni, is a fruit that is commonly used as a dietary supplement because of its antioxidant, anti-inflammatory and hepatoprotective properties. Many dietary supplements can cause inhibition or induction in the Cytochrome P450 system. The effects of the Morinda citrifolia in the CYP4503A4 and CYP4502C9 of Chinese Hamster Ovarian Cells (CHO cells) was studied. CHO cells were cultivated and treated with the Noni’s extracts by a period of 48 hours. Three extracts were prepared: a pulp extract, an organic extract of Noni’s skin and an aqueous skin extract. Using an appropriate luminescent compound the luminescence of each sample was determined. Cytochrome P450 activity was determined. Results showed that the enzyme activity was affected. For the CYP3A4 the control group enzyme activity was of 0.0679 pmol of Luciferin/ pmol of CYP3A4/ minutes and for the experimental groups 0.0609 pmol of Luciferin/ pmol of CYP3A4/ minutes (pulp), 0.0581 pmol of Luciferin/ pmol of CYP3A4/ minutes (organic) and 0.0654 pmol of Luciferin/ pmol of CYP3A4/ minutes (aqueous). For the CYP2C9 the results were: 0.0858 pmol of Luciferin/ pmol of CYP2C9/ minutes for the control group, 0.0834 pmol of Luciferin/ pmol of CYP2C9/ minutes for the pulp, and for the skin’s extracts: 0.0754 pmol of Luciferin/ pmol of CYP2C9/ minutes (organic) and 0.0788 pmol of Luciferin/ pmol of CYP2C9/ minutes (aqueous). The results showed a reduction of CYP3A4 enzyme activity when treated with the organic extract of Noni’s skin, meaning it significally affects the isoenzyme activity. Noni’s pulp and aqueous extract does not significally affect CYP3A4.
